TarD provides a pool of CDP-activated glycerol-3-phosphate for TarB to use for UDP-disaccharide glycerophosphate priming. Wall teichoic acids in S. aureus use ribitol exclusively as their polyol extension monomer, therefore WTA enzymes in S. aureus are assigned the "tar" nomenclature. In contrast, S. epidermidis uses glycerol as its polyol extension monomer so orthologous enzymes in S. epidermidis are assigned the "tag" nomenclature. The utilization of glycerol phosphate by TarB and TarD for polyol priming further blurs the line between the "Tar" and the "Tag" nomenclature but because the polymerizing polyol will be ribitol, the enzymes retain the "Tar" nomenclature.
