mnmL : tRNA 5-carboxymethylaminomethyl-2-thiouridine(34) oxidative decarboxylase MnmL [1]

Transfer RNAs containing uridine at wobble anticodon position 34 often require modifications to provide maximal fidelity during protein synthesis starting from a shared 5-carboxymethylaminomethyl-2-thiouridine(34) (“cmnm5s2U”, “$”) generated by MnmAEG. One such modification involves SAM-dependent oxidative decarboxylation of 5-carboxymethylaminomethyl-2-thiouridine(34) to generate 5-aminomethyl-2-thiouridine(34) and subsequent SAM-dependent methylation to 5-methylaminomethyl-2-thiouridine(34) by the MnmLM complex. A small amount of 5-aminomethyl-2-thiouridine(34) can also be provided by MnmEG using ammonium as a substrate. These two enzymatic activities are shared in the bifunctional enzyme MnmC in gram-negative bacteria.
