The type IV restriction endonuclease SauUSI binds to to DNA containing a 5'-S5mCNGS-3' target recognition domain. Its cleavage mechanism is complex involving lateral translocation along the DNA accompanied by ATP hydrolysis (analogous to type III restriction endonucleases) but preferential DNA cleavage only when stationary and "hit" by an actively translocating enzyme. This results in cleavage near target recognition domains at low enzyme concentrations due to stationary enzymes bound at TRDs and shredding of DNA at high enzyme concentrations as translocating enzymes bump into each other, stop their translocation, and engage their DNA cleavage as actively-translocating enzymes behind them "pile up".
