Staphylococcal prophage capsids are typically assembled from major capsid proteins and scaffold proteins with scaffold proteins being lost during capsid maturation. Serogroup A capsids incorporate both functionalities within the same protein with the δ domain performing the scaffolding activity. 535 MCP monomers are involved in the assembly starting from a nucleus of 5 hexamers, extended into a prolate capsid by cylindrical arrangements of six rings of ten hexamers. After assembly, the δ scaffolding domain is cleaved by a crotonase-like head maturation protease between lysine-128 and leucine-129 generating a mature capsid.
