Transfer of diglucosyl-diacylglycerol from the inner leaflet of the cell membrane to the outer leaflet by diglucosyl-diacylglycerol flippase is the second committed step in teichoic acid synthesis. This activity is primarily regulated by binding of the coordination of zonal exclusion proteins CozEa and CozEb which inhibit the flippase activity. All subsequent lipoteichoic acid biosynthesis and modification steps will occur in the extracellular environment.
